Product Introduction:
RNAfixer is an aqueous, non-toxic tissue preservation reagent that can quickly penetrate into the cytoplasm of fresh tissue cells, stabilizing and protecting intracellular RNA in situ under non-frozen conditions. Immediately immersing tissue sections into RNAfixer after collection does not affect the quality and quantity of RNA extracted later. RNAfixer eliminates the inconvenience of needing to process RNA samples immediately or store them in liquid nitrogen.

After immersion in RNAfixer, RNA in fresh tissue cells can be well preserved at 37°C for 1 day, at 25°C for 1 week, at 4°C for 1 month, and long-term at -20°C or -80°C. RNA virus samples (such as HCV and HIV) can be preserved at 37°C for 1 month.

Product Features:
1.Easy to operate: Cut the tissue into appropriate sizes and immerse in RNAfixer to prevent RNA degradation.
2.No liquid nitrogen required: Enables sample preservation without liquid nitrogen, dry ice, or a -80°C refrigerator, especially suitable for rapid and large-scale collection of clinical and field samples.
3.Convenient transportation: Treated samples can be stored at 25°C for 1 week, making sample mailing and transportation easier and cheaper, facilitating academic cooperation and exchange.
4.Multiple freeze-thaw cycles: Samples treated with RNAfixer can undergo repeated freeze-thaw cycles, and various treatments can be performed on the samples without affecting the quality of the finally extracted RNA.
5.High comparability: RNAfixer can reduce errors in large-scale sample processing and increase the comparability between experimental data, which is particularly useful for large-scale gene expression profile analysis.
6.Wide compatibility: A variety of total RNA extraction reagents can be used to extract samples preserved in RNAfixer. It can also be directly used for tissue sectioning, immunology, and flow cytometry analysis without affecting the quality of RNA extraction.

Application Scope:
Suitable for animal tissues (heart, liver, kidney, muscle, testis, brain, spleen, etc.), cultured cells, RNA viruses, Drosophila, bacteria, leukocytes, whole blood, and some plant tissues.

Extraction Methods of Samples in RNAfixer:
Take the sample out of RNAfixer. RNAfixer can be directly poured into the sink and rinsed with tap water without special treatment.

1.Tissues:Use clean tweezers to take the sample out of RNAfixer, blot the residual RNAfixer with absorbent paper, grind with liquid nitrogen like fresh tissue, and then homogenize with Trizol (Catalog No.: 2101B) to extract RNA.

2.Cells:There are two options for cells stored in RNAfixer: one is to add 10 times the volume of Trizol (Catalog No.: 2101B) directly to the mixture of cells and RNAfixer for extraction; the other is to centrifuge to pellet the cells, remove RNAfixer, and then extract RNA with Trizol (Catalog No.: 2101B).
a.RNA Extraction After Removing RNAfixer:Cells stored in RNAfixer become less fragile and can withstand higher centrifugal speeds without lysis. We have experience in successfully collecting cells by centrifugation at 5000 g. Since the strength of each type of cell is different, a pre-experiment with unimportant cells is recommended to ensure that centrifugation at the used speed will not damage the cells. Another option is to dilute the mixture of RNAfixer and cells with an equal volume of PBS before centrifugation to reduce the density of the solution, allowing the cell suspension to pellet.
b.Direct RNA Extraction Without Removing RNAfixer:Alternatively, directly add 10 times the volume of one-step extraction reagent (such as Trizol, TRI reagent) to the mixture of cells and RNAfixer, then follow the normal steps for operation.
